If I were casting it, I would break up the trio (almost foursome) and cast each individually and I'd give
each the opportunity to reinvent the character. (Strangelove not necessarily wheelchair bound, for example.)
(Although, the Depp solution is a great one.)

Choices in no particular order:

Major King Kong: Randy Quaid

Brig. Gen. Jack D. Ripper: Burt Reynolds
Robert De Niro
(I really want to say Jack Nicholson here.)

General Turgidson: Fred Ward
Michael Gross

Muffley: Warren Beatty
Harrison Ford

Strangelove: Robin Williams
Johnny Depp

Mandrake: Hugh Laurie
Pierce Brosnan

COL Guano: William H. Macy
